Acute myeloid leukemia (AML) is a heterogeneous disease with poor clinical outcomes. We have previously shown that constitutive activation of NADPH oxidase 2 (NOX2), resulting in over-production reactive oxygen species (ROS), occurs over 60% AML patients. also increased ROS production promotes glucose uptake and proliferation cells, mediated by changes carbohydrate metabolism. Given carbohydrate, lipid, protein metabolisms are all intricately interconnected, we aimed to examine the effect cellular levels on these pathways establish further evidence rewires metabolism AML. carried out metabolomic profiling cell lines which NOX2-derived was inhibited conversely cells treated exogenous H O . report significant ROS-specific metabolic alterations sphingolipid metabolism, fatty acid oxidation, purine amino homeostasis glycolysis. These data provide directed potential for targeting as novel therapeutic arm combat this disease.
